The matrix metalloproteinases (MMP) are a family of peptidase enzymes responsible for the degradation of extracellular matrix components, including collagen, gelatin, fibronectin, laminin and proteoglycan. Transcription of MMP genes is differentially activated by phorbol ester, lipopolysaccharide (LPS) or staphylococcal enterotoxin B (SEB). MMP catalysis requires both calcium and zinc. MMP-3, MMP-10 and MMP-11 (also designated stromelysin-1, 2 and 3, respectively) activate procollagenase. MMP-3 activation of procollagenase can occur via two pathways. Direct activation by MMP-3 is slow and activation by MMP-3 in conjunction with tissue or plasma proteinases is rapid. MMP-10 is expressed in small intestine, and at lower levels in lung and heart. MMP-11 is specifically expressed in stromal cells of breast carcinomas and contributes to epithelial cell malignancies. Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI 6. 0) subfamilies. This antibody recognizes acidic (Type I or LMW) and basic (Type II or HMW) cytokeratins, including 59 kDa (CK4); 58 kDa (CK5); 56 kDa (CK6); 52 kDa (CK8); 56. 5 kDa (CK10); 53 kDa (CK13) and 45 kDa (CK18). This is a broad-spectrum antibody, which has been reported to differentiate epithelial tumors from non-epithelial tumors. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. This antibody recognizes proteins of 31-44 kDa, which are identified as Clathrin Light Chains (both A & B). Clathrin is composed of three heavy chains and three light chains, which associate non-covalently to form a triskelion structure. Clathrin light chain regulates the self-assembly of triskelions onto intracellular membranes. Clathrin light chain subunits (LCA and LCB) contribute to regulation of coated vesicle formation to sort proteins during receptor-mediated endocytosis and organelle biogenesis. LCA and LCB are encoded by two discrete genes. They share only 60% homology, and have certain features in common. Alpha-1 Antichymotrypsin (AACT) is a plasma protease inhibitor synthesized in the liver as a single glycopeptide chain. In human, the normal serum level of AACT is about one-tenth that of alpha-1-antitrypsin (AAT), with which it shares nucleic acid and protein sequence homology. Both are major acute phase reactants; their concentrations in plasma increase in response to trauma, surgery and infection. Elevated levels of AACT are widely, but not universally, reported in the cerebrospinal fluid and plasma of AD patients. Prostate-specific antigen (PSA) and its SDS-stable complex with AACT are in widespread use as markers for the diagnosis of prostate cancer. AACT deficiency may also be a possible cause of chronic liver disease. AACT antibody reacts with histiocytes and histiocytic neoplasms. CD9 is a type IV transmembrane glycoprotein with four transmembrane domains. CD9 on pre-B cells may play a role in cell-cell adhesion. In addition, CD9 may play a role in signal transduction mediated by interaction with low molecular weight GTP binding proteins. CD9 is expressed on early B cells, eosinophils, basophils and activated T cells and is a major component of the platelet cell surface. This antibody recognizes a protein of 130 kDa, identified as Ksp-cadherin. Cadherins form a superfamily of related glycoproteins that mediate calcium-dependent cell adhesion and transmit signals from the extracellular matrix to the cytoplasm. Cadherins have been implicated in embryogenesis, tissue morphogenesis, tissue structure maintenance, cell polarization, neoplastic invasiveness and metastasis, and membrane transport. It is suggested that Ksp-cadherin is a marker for terminal differentiation of the basolateral membranes of renal tubular epithelial cells. Within the kidney, Ksp-Cadherin is found exclusively in the basolateral membrane of renal tubular epithelial cells and collecting duct cells, and not in glomeruli, renal interstitial cells, or blood vessels. The progression of cells through the cell cycle is regulated by a family of proteins designated cyclin-dependent kinases (Cdks). Sequential activation of individual members of this family and their consequent phosphorylation of critical substrates promotes orderly progression through the cell cycle. This MAb reacts with a CD32 (FcgRII) epitope distinct from that defined by MAb 8.26 and the epitope overlaps with that of MAb 7.30 (cluster 4). It displays a stronger reaction with Daudi than with U937 cells. The epitope is located in domain 2 of FcgRIIa. Its Fab'2 fragments block immune complex binding. CD32 (FcgRII) is a type 1 transmembrane glycoprotein that mediates several functions including phagocytosis, cytotoxicity, and immunomodulation as well as platelet aggregation. Three genes (A, B, and C) encode CD32 and at least 6 isoforms are generated via alternative mRNA splicing, i.e., IIa1, IIa2, IIb1, IIb2, IIb3 and IIc. The major constituents of mucus, the viscous secretion that covers epithelial surfaces such as those in the trachea, colon, and cervix, are highly glycosylated proteins called mucins. These glycoproteins play important roles in the protection of the epithelial cells and have been implicated in epithelial renewal and differentiation. This gene encodes an integral membrane glycoprotein found on the cell surface, although secreted isoforms may exist. MUC-4 transcripts have been detected in normal respiratory epithelium and lung. MUC-4 is a very specific (100%) and sensitive (90%) marker of lung adenocarcinomas and is negative for mesotheliomas. Reportedly, MUC-4 expression in invasive ductal carcinoma of the pancreas is an independent factor for poor prognosis. The BRCA 1 Associated Protein 1 (BAP1) protein belongs to the ubiquitin C-terminal hydrolase subfamily of de-ubiquitination enzymes that are involved in the removal of ubiquitin from proteins. The encoded enzyme binds to the breast cancer type 1 susceptibility protein (BRCA1) via the RING finger domain of the latter and acts as a tumor suppressor.
